Malate exchange between the cytosol and mitochondria.

چکیده
1. By comparing the relative isotopic yields in glucose and CO(2) from precursors of mitochondrial and cytosolic malate, it is evident that the rate of isotopic exchange between these compartments is rapid. 2. A variety of potential inhibitors of malate exchange were tested, but no specific and effective inhibitor of the isotopic exchange has been found. 3. Compounds such as n-butylmalonate and p-iodobenzylmalonate, which have been used as inhibitors of the malate-phosphate transport system in isolated mitochondria, do not appear to be sufficiently specific to be useful in studies with intact cells.

Oxaloacetate and malate transport by plant mitochondria.
The permeability of mitochondria from pea (Pisum sativum L. var Kleine Rheinländerin) leaves, etiolated pea shoots, and potato (Solanum tuberosum) tuber for malate, oxaloacetate, and other dicarboxylates was investigated by measurement of mitochondrial swelling in isoosmolar solutions of the above mentioned metabolites. For the sake of comparison, parallel experiments were also performed with r...
متن کاملInhibition of gluconeogenesis by butylmalonate in perfused rat liver.
The role of mitochondrial anion exchange reactions in the control of gluconeogenesis in perfused livers from fasted rats was investigated by using Z-n-butylmalonate to inhibit malate transport across the mitochondrial membrane. Glucose production from pyruvate or lactate was inhibited 50% to 80% by 5 m&r butylmalonate. Oleate (0.5 or 1.5 mr+r) increased glucose production from pyruvate from 82 ...
متن کاملAcute and Chronic Ethanol Treatment in Viuo Increases Malate-Aspartate Shuttle Capacity in Perfused Rat Liver*
The effects of acute and chronic treatment with ethanol on transport of reducing equivalents into mitochondria via the malate-aspartate shuttle were studied in perfused rat liver. The shuttle capacity was estimated from the decrease in rates of glucose production from the reduced substrate sorbitol caused by an increase in the NADH/NAD+ ratio in the cytosol due to metabolism of ethanol. The gre...
متن کاملFactors affecting the kinetics and equilibrium of exchange reactions of the citrate-transporting system of rat liver mitochondria.
Benzene 1,2,3-tricarboxylic acid was found to be a sufficiently potent inhibitor of citrate transport by rat liver mitochondria that it may be used in “inhibitor stop” experiments. It was thus possible to measure the initial rates of exchange reactions catalyzed by the citrate-transporting system and to study the dependence of the rate on temperature and the concentrations of citrate, malate, a...
متن کاملThe oxoglutarate/malate carrier of rat brain mitochondria operates by a uniport exchange mechanism.
Here, the oxoglutarate carrier, already isolated from various sources and described in the literature, has been purified from rat brain and reconstituted in proteoliposomes for an accurate kinetic study. The rate of uptake of labelled oxoglutarate and malate has been measured in various conditions, essentially in double substrate experiments. The data so obtained fit the hypothesis that the car...
